2 SLI or Crossfire video cards will always run as fast as or even faster than one video card.
But without Direct X 12, multi-gpu setup can't do what it promised on ad (i.e 2 RX480 > 1 GTX 1080).
AMD is nice to keep 3 way and 4 way crossfire alive, unlike nVidia GTX 1070/1080 only supports 2 way SLI.
If I am in your shoes and have $$, I would go with X99, not Z170.
X99 allows way more PCI Express lanes.
If you plan to use PCI Express SSD like the Intel 750 series, then that means you need another PCI Express x4 speed or x16 long slot.
A Z170 board can have at most, a total of 4 PCI Express x16 slots, when all 4 slots are utilized, they run at x8, x4, x4, x4 speed.
The cheapest Z170 board has only 2 PCI Express x16 slots (run at x16 and x4 speed only, can't be modified). These boards may not meet your requirements.
And buying an expensive Z170 board makes little sense when the cost of i7-6700K and the cost of Z170 board together will be nearly as expensive as a cheapest X99 setup. You can get at least 6 cores and at least 28 PCI Express lanes on a X99 setup.
Use newegg.com and select the PCI Express slots to show the motherboards that meet your requirements.
I think you will need at least 3 PCI Express x16 slots, x8 speed for rx480, x4 for rx480, x4 for PCI Express interface SSD.
